It &#105;&#115; swallowed, injected, &#111;&#114; snorted. Ketamine &#097;&#108;&#115;&#111; leads &#116;&#111; problems with thinking, memory, and concentration. </p>
<p>A &#110;&#101;&#119; study &#102;&#111;&#108;&#108;&#111;&#119;&#101;&#100; &#111;&#118;&#101;&#114; 1,200 people who &#104;&#097;&#100; &#117;&#115;&#101;&#100; ketamine in &#116;&#104;&#101; last year. Of &#116;&#104;&#101; users, 17 percent were addicted &#116;&#111; it, and 26 percent of users &#104;&#097;&#100; experienced urinary problems, including pain in &#116;&#104;&#101; &#108;&#111;&#119;&#101;&#114; abdomen, burning &#111;&#114; stinging during urination, bloody urine, and incontinence (urine leakage).</p>
<p>The severity of urinary symptoms was connected &#116;&#111; how often and &#116;&#104;&#101; size of &#116;&#104;&#101; dose of ketamine &#116;&#104;&#101; participants &#117;&#115;&#101;&#100;. </p>

<p>Ketamine &#105;&#115; known &#116;&#111; cause irritation and inflammation of &#116;&#104;&#101; mucous membranes, &#097;&#115; &#119;&#101;&#108;&#108; &#097;&#115; thickening of &#116;&#104;&#101; bladder wall and kidney problems. &#098;&#117;&#116; &#116;&#104;&#101; exact mechanisms by &#119;&#104;&#105;&#099;&#104; ketamine &#097;&#099;&#116;&#117;&#097;&#108;&#108;&#121; &#099;&#097;&#117;&#115;&#101;&#115; these changes in &#116;&#104;&#101; urinary tract are &#110;&#111;&#116; &#119;&#101;&#108;&#108; understood. </p>

<p>A new international study has &#109;&#097;&#100;&#101; a discovery &#116;&#104;&#097;&#116; &#109;&#097;&#121; reshape the way  schizophrenia &#105;&#115; treated.</p>
<p>Some &#116;&#119;&#111; dozen researchers &#102;&#114;&#111;&#109; University &#111;&#102; California San Diego School &#111;&#102; Medicine, Trinity College Dublin &#097;&#110;&#100; &#101;&#108;&#115;&#101;&#119;&#104;&#101;&#114;&#101; have identified a gene mutation strongly linked &#116;&#111; schizophrenia &#116;&#104;&#097;&#116; &#109;&#097;&#121; &#098;&#101; an important new target &#102;&#111;&#114; the development &#111;&#102; drug therapies.</p>
<p>Schizophrenia &#105;&#115; a chronic, severe &#097;&#110;&#100; disabling brain disorder, &#119;&#105;&#116;&#104; symptoms &#116;&#104;&#097;&#116; include hallucinations, delusions &#097;&#110;&#100; &#116;&#104;&#111;&#117;&#103;&#104;&#116; disorders. It is believed &#116;&#111; &#098;&#101; caused &#098;&#121; environmental &#097;&#110;&#100; genetic factors, especially the &#108;&#097;&#116;&#116;&#101;&#114;: the illness occurs in 1 percent &#111;&#102; the general population, but in 10 percent &#111;&#102; people who have a first-degree relative &#119;&#105;&#116;&#104; the disorder, &#115;&#117;&#099;&#104; as a parent or sibling.</p>
<p>Current therapies are only partially effective &#119;&#105;&#116;&#104; &#108;&#105;&#116;&#116;&#108;&#101; progress being &#109;&#097;&#100;&#101; in identifying effective new treatments &#111;&#118;&#101;&#114; several decades.</p>
<p>In the last three years, researchers have discovered &#116;&#104;&#097;&#116; rare mutations &#097;&#116; &#109;&#097;&#110;&#121; locations in the human genome resulted in significantly higher risk &#111;&#102; schizophrenia.</p>
<p>These mutations consisted &#111;&#102; copy number variants or CNVs ?a type &#111;&#102; genetic variation in which the number &#111;&#102; copies &#111;&#102; a gene differs between individuals. The findings &#119;&#101;&#114;&#101; the first conclusive evidence &#116;&#104;&#097;&#116; rare mutations &#099;&#097;&#110; cause schizophrenia, but this did not identify the specific genes involved.</p>
<p>In the current study, researchers scanned &#102;&#111;&#114; CNVs in the genomes &#111;&#102; 8,290 individuals &#119;&#105;&#116;&#104; diagnosed cases &#111;&#102; schizophrenia &#097;&#110;&#100; 7,431 healthy controls.</p>
<p>The study confirmed CNVs identified in earlier studies, but uncovered an important new finding: duplications &#097;&#116; the tip &#111;&#102; chromosome 7q &#119;&#101;&#114;&#101; detected in individuals &#119;&#105;&#116;&#104; schizophrenia &#097;&#116; a rate 14 times higher than in healthy individuals. &#116;&#104;&#101;&#115;&#101; duplications impact a gene coding &#102;&#111;&#114; the brain receptor VIPR2.</p>
<p>Formally known as the Vasoactive Intestinal Peptide Receptor 2, VIPR2 &#105;&#115; expressed in the nervous system, including in the brain, blood vessels &#097;&#110;&#100; gastrointestinal tract.</p>
<p>Previous studies have shown &#116;&#104;&#097;&#116; VIPR2 helps &#116;&#111; regulate the formation &#097;&#110;&#100; activity &#111;&#102; neurons in the brain. In mice, VIPR2 &#097;&#108;&#115;&#111; has &#098;&#101;&#101;&#110; found &#116;&#111; play important roles in behavioral processes, including learning &#097;&#110;&#100; timing &#111;&#102; daily activity. The study &#110;&#101;&#120;&#116; measured expression &#111;&#102; the VIPR2 gene in blood cells &#102;&#114;&#111;&#109; patients, &#116;&#104;&#101;&#121; found &#116;&#104;&#097;&#116; individuals &#119;&#105;&#116;&#104; mutations had greater expression &#111;&#102; VIPR2 &#097;&#110;&#100; greater activity &#111;&#102; the receptor.</p>
<p>&#8220;This suggests &#116;&#104;&#097;&#116; the mutations increase signaling in the Vasoactive Intestinal Peptide pathway,&#8221; according &#116;&#111; co-author Dr. Aiden Corvin.</p>

<p>Schizophrenia &#105;&#115; a chronic, severe and disabling brain disorder, with symptoms &#116;&#104;&#097;&#116; include hallucinations, delusions and thought disorders. &#105;&#116; &#105;&#115; believed to be caused &#098;&#121; environmental and genetic factors, &#109;&#111;&#115;&#116; notably &#116;&#104;&#101; &#108;&#097;&#116;&#116;&#101;&#114;: &#116;&#104;&#101; illness occurs &#105;&#110; 1 percent of &#116;&#104;&#101; general population, or 10 percent of people who &#104;&#097;&#118;&#101; a first-degree relative with &#116;&#104;&#101; disorder, such &#097;&#115; a parent or sibling. </p>
<p>In previous work, Sebat and collaborator Mary-Claire King, a professor of medical genetics &#097;&#116; &#116;&#104;&#101; University of Washington, discovered &#116;&#104;&#097;&#116; rare mutations &#097;&#116; &#109;&#097;&#110;&#121; locations &#105;&#110; &#116;&#104;&#101; human genome resulted &#105;&#110; significantly higher risk of schizophrenia. &#116;&#104;&#101;&#115;&#101; mutations consisted of copy number variants or CNVs &ndash; a type of genetic variation &#105;&#110; which &#116;&#104;&#101; number of copies of a gene differs between individuals. &#116;&#104;&#101; findings were &#116;&#104;&#101; first conclusive evidence &#116;&#104;&#097;&#116; rare mutations &#099;&#097;&#110; cause schizophrenia, but they &#100;&#105;&#100; not identify &#116;&#104;&#101; specific genes involved.</p>
<p>The latest study goes &#109;&#117;&#099;&#104; &#102;&#117;&#114;&#116;&#104;&#101;&#114;. Researchers scanned for CNVs &#105;&#110; &#116;&#104;&#101; genomes of 8,290 individuals with diagnosed cases of schizophrenia and 7,431 healthy controls. &ldquo;We &#102;&#111;&#117;&#110;&#100; very strong links to multiple sites &#105;&#110; &#116;&#104;&#101; genome,&rdquo; &#115;&#097;&#105;&#100; Sebat. &ldquo;Some had &#098;&#101;&#101;&#110; picked &#117;&#112; before &#105;&#110; earlier studies, but we uncovered a very &#105;&#109;&#112;&#111;&#114;&#116;&#097;&#110;&#116; &#110;&#101;&#119; finding: duplications &#097;&#116; &#116;&#104;&#101; tip of chromosome 7q were detected &#105;&#110; individuals with schizophrenia &#097;&#116; a rate14 times higher than &#105;&#110; healthy individuals. &#116;&#104;&#101;&#115;&#101; CNVs impact a gene &#116;&#104;&#097;&#116; &#105;&#115; &#105;&#109;&#112;&#111;&#114;&#116;&#097;&#110;&#116; for brain development &ndash; &#116;&#104;&#101; neuropeptide receptor VIPR2.&rdquo;</p>
<p>Formally &#107;&#110;&#111;&#119;&#110; &#097;&#115; &#116;&#104;&#101; Vasoactive Intestinal Peptide Receptor 2, VIPR2 &#105;&#115; expressed &#105;&#110; &#116;&#104;&#101; nervous &#115;&#121;&#115;&#116;&#101;&#109;, including &#105;&#110; &#116;&#104;&#101; brain, blood vessels and gastrointestinal tract. Previous studies &#104;&#097;&#118;&#101; shown &#116;&#104;&#097;&#116; VIPR2 helps to regulate &#116;&#104;&#101; formation and activity of neurons &#105;&#110; &#116;&#104;&#101; brain. &#105;&#110; mice, VIPR2 &#097;&#108;&#115;&#111; &#104;&#097;&#115; &#098;&#101;&#101;&#110; &#102;&#111;&#117;&#110;&#100; to play &#105;&#109;&#112;&#111;&#114;&#116;&#097;&#110;&#116; roles &#105;&#110; behavioral processes, including learning and timing of daily activity. </p>

<p>&#8220;Take two aspirin &#097;&#110;&#100; call me in &#116;&#104;&#101; morning.&#8221;  For schizophrenia?  &#112;&#101;&#114;&#104;&#097;&#112;&#115;.  New research suggests that aspirin may be &#111;&#102; benefit in reducing &#116;&#104;&#101; symptoms &#111;&#102; schizophrenia.</p>
<p>Schizophrenia is a &#115;&#101;&#114;&#105;&#111;&#117;&#115; psychological disorder characterized by impairments in thinking that &#099;&#097;&#110; lead to delusions, difficulty in determining reality, &#097;&#110;&#100; hallucinations.  Sufferers are &#097;&#116; high risk for substance abuse and suicide.  &#117;&#112; to 2.2 million Americans suffer from &#116;&#104;&#101; disease. </p>
<p>There is &#110;&#111; single &#107;&#110;&#111;&#119;&#110; cause for schizophrenia, but chemical &#097;&#110;&#100; structural abnormalities &#111;&#102; &#116;&#104;&#101; brain as well as genetics appear to play a role.  Treatments include antipsychotics &#097;&#110;&#100; psychosocial therapy.</p>
<p>Prior research noted an improvement &#111;&#102; symptoms in schizophrenia when patients were &#116;&#097;&#107;&#105;&#110;&#103; anti-inflammatories &#115;&#117;&#099;&#104; &#097;&#115; celexicob, a cyclooxygenase-2 inhibitor.  &#116;&#104;&#101;&#115;&#101; results suggest that inflammation may play a role in causing some &#111;&#102; &#116;&#104;&#101; symptoms &#111;&#102; schizophrenia.</p>
<p>Dr. Laan Grobbee &#097;&#110;&#100; &#104;&#105;&#115; team &#111;&#102; researchers in &#116;&#104;&#101; Netherlands evaluated whether &#117;&#115;&#105;&#110;&#103; a medication to treat inflammation might improve &#116;&#104;&#101; symptoms &#111;&#102; schizophrenia.  They used aspirin &#097;&#115; an anti-inflammatory because cyclooxygenase-2 inhibitors are &#107;&#110;&#111;&#119;&#110; to increase &#116;&#104;&#101; risk &#111;&#102; cardiovascular disease.</p>
<p>Grobbee conducted a randomized, double-blinded, placebo-controlled study with 80 patients &#119;&#104;&#111; &#104;&#097;&#100; had schizophrenia for &#102;&#101;&#119;&#101;&#114; &#116;&#104;&#097;&#110; &#102;&#105;&#118;&#101; years.  Patients were randomly assigned to either 1000mg &#111;&#102; aspirin or placebo, &#097;&#110;&#100; treated for three months.</p>
<p>Patients &#119;&#104;&#111; required antipsychotic medications (70 percent) continued to be treated with olanzapine, clozapine, or risperidone.  All patients were given a proton-pump inhibitor as a precaution to protect them from any &#112;&#111;&#115;&#115;&#105;&#098;&#108;&#101; stomach irritation due to the aspirin.</p>
<p>The Posititive &#097;&#110;&#100; Negative Syndrome Scale (PANSS) was used to measure &#116;&#104;&#101; change in symptoms &#097;&#116; &#116;&#104;&#101; beginning &#111;&#102; &#116;&#104;&#101; treatment &#097;&#110;&#100; &#097;&#102;&#116;&#101;&#114; three months.</p>
<p>After three months, &#116;&#104;&#101; patients treated with aspirin &#115;&#104;&#111;&#119;&#101;&#100; a significant reduction in &#116;&#104;&#101; PANSS (4.86 points) compared to &#116;&#104;&#101; patients treated with placebo (1.57 points). </p>
<p>Cognitive function was &#097;&#108;&#115;&#111; assessed, &#097;&#110;&#100; &#116;&#104;&#101;&#114;&#101; &#100;&#105;&#100; &#110;&#111;&#116; appear to be any negative effects from &#116;&#104;&#101; aspirin.</p>
<p>These results suggest that &#116;&#104;&#101;&#114;&#101; may indeed be an inflammatory component &#111;&#102; schizophrenia that could &#112;&#101;&#114;&#104;&#097;&#112;&#115; be responsive to anti-inflammatory agents &#115;&#117;&#099;&#104; &#097;&#115; aspirin.  Grobbee noted an even greater improvement in patients with altered immune function.</p>
<p>The authors suggest that aspirin may &#097;&#108;&#115;&#111; theoretically work in treatment &#111;&#102; schizophrenia through other mechanisms &#115;&#117;&#099;&#104; &#097;&#115; &#116;&#104;&#101; N-methyl d-aspartate receptor, &#097;&#110;&#111;&#116;&#104;&#101;&#114; active area &#111;&#102; research.</p>
<p>Given &#114;&#101;&#099;&#101;&#110;&#116; research &#115;&#104;&#111;&#119;&#105;&#110;&#103; schizophrenics &#097;&#116; increased risk for cardiovascular disease &#097;&#110;&#100; other high-risk metabolic conditions &#115;&#117;&#099;&#104; &#097;&#115; defects in insulin function, aspirin may be &#111;&#102; benefit in more &#116;&#104;&#097;&#110; &#111;&#110;&#101; &#119;&#097;&#121;.</p>
